Gly246Val mutation introduces a bulky valine side chain at the gatekeeper position, sterically hindering compound entry into the ATP-binding cleft. Model fit quality reduced vs. WT (R² = 0.995 vs. 0.999), suggesting potential minor population of alternative binding modes.
